How Ion Exchange Chromatography Works
Ion exchange (IEX) chromatography separates charged molecules based on their reversible electrostatic interaction with ionic functional groups bonded to the silica surface. Typical analytes include:
Organic bases

Proteins and peptides
Other positively charged or polar compounds
Retention occurs through the exchange of ions between the analyte in the mobile phase and the oppositely charged ligands on the stationary phase. Retention and selectivity can be adjusted by modifying mobile phase pH and ionic strength.
The functional pH range for Cogent IEX SCX™ columns is 2.5 to 8.
Strong Cation Exchange (SCX) Selectivity
Cation exchange columns contain negatively charged functional groups that retain positively charged (basic) analytes.
Cogent IEX SCX™ columns utilize sulfonic acid functional groups, which provide strong and consistent ionic interactions with basic compounds. Because these functional groups remain ionized across the usable pH range, retention behavior is stable and predictable.
What “Strong” Ion Exchange Means
The term “strong” ion exchange does not describe stronger binding to analytes. Instead, it refers to the ionization behavior of the functional group:
Strong cation exchange (SCX): The functional group is always ionized, regardless of mobile phase pH.
This constant charge state simplifies method development and ensures reproducible performance across different operating conditions.
